Overview of Sunifiram 

Sunifiram is an Ampakine drug (acts via AMPA receptors). It is a synthetic derivative of Piracetam and has a somewhat similar chemical structure. Still, despite having comparable anti-amnesic effects, Sunifiram demonstrates significantly higher potency than piracetam in preclinical models.

Sunifiram was originally developed to treat the symptoms of cognitive decline in Alzheimer’s disease patients. However, due to the lack of funding, no human trials were conducted to investigate its effects. All the evidence about its effects comes from animal studies.

Furthermore, it has been reported that Sunifiram attenuates experimentally induced memory deficits in research models. It alleviates cognitive deficits and possesses potent anti-amnesic activity in preclinical models. Working Mechanism of Sunifiram?

The exact working mechanism of Sunifiram remains unknown, but it has been shown to:

Stimulate the release of acetylcholine

In one animal study, Sunifiram reportedly increased acetylcholine release. Another preclinical study showed it blocked the effects of scopolamine, a drug that blocks acetylcholine release.

Activate AMPA receptors

Study reports suggest that unifiram appears to modulate glutamatergic signaling involving NMDA and AMPA receptor pathways. Plus, it may block AMPA receptor activation by drugs that suppress the activity of AMPA receptors.

Regulate glucose uptake

A cell-based study reports that Sunifiram may increase glucose uptake at a lower dose and increase it at a mega dose.

Activate NDMA glutamate receptors.

Another cell-based study suggests how Sunifiram may activate NDMA glutamate receptors. That is, by promoting the release of the neurotransmitter glutamate, which is proven to bind to glutamate receptors.

In research models, exposure to Sunifiram was linked with improved performance in tasks that measure learning and recognition, including passive avoidance and social recognition tests. A few studies have reported additional neurological effects of Sunifiram. Some experimental findings report mild effects on heat-related pain responses and changes in sleep-related behavior in research models.
